ABSTRACT

Objective To compare the clinical effect of vascularized fibular flap transplantation and vancomycin sulfate calcium on chronic osteomyelitis of the tibia and bone defects. Methods A total of 35 cases with chronic tibial osteomyelitis and bone defect were involved and divided into group A with 21 cases receiving anastomosis of vascularized fibular flap transplantation and group B with 14 cases recciving vancomycin sulfate calcium.Infection control rate and Enneking score were compared in both groups.Simultaneous detection of two groups preoperative, postoperative 2 weeks,1 month and 3 months of the levels of CRP and PCT were conducted. Results Postoperative CRP and PCT were under control in both groups. No statistical significance was found in the comparison of CRP and PCT preoperative and postoperative 2 weeks,1 month and 3 months. In terms of infection control rate, two groups had no signiticant difference (P > 0.05) and there was statistical significance of the comparison of postoperative Enneking score (P < 0.05).Conclusions The clinical results of the two methods are satisfied. The infection degree and the length of the bone defect are decisive factors in choosing the treatment method and sometimes the combination of two treatment methods may be a better choice.

ABSTRACT

Aim To investigate the effect of Ding''s herb enema prescription on intestinal tissue related target in rat colitis induced by dextran sulfate sodium(DSS), and to elucidate the mechanism of Ding''s herb enema prescription in improving the intestinal inflammation and intestinal fibrosis. Methods Rats were fed with 3.5% DSS.The rats were randomly divided into positive drug group, model group, Control group, and Ding''s herb enema prescription group. The positive drug group was treated with mesalazine enema, and Ding''s herb enema prescription group was treated with Ding''s herb enema prescription. The colon mucosa was taken once a day for 6 weeks. The changes of intestinal inflammatory response and intestinal fibrosis related proteins were detected by GSR-CAA-67 antibody protein array, and the differentially expressed proteins were screened out. Results Eight proteins showed statistical differences, including IFN-γ, erythropoietin(EPO), TIMP-2, TIM-1, IL-6, TIMP-1, TNF-α, IL-22 (P0.05).Conclusions Ding''s herb enema prescription has the effect of multiple targets, which may improve the intestinal inflammatory response and intestinal fibrosis to achieve the purpose of treatment of ulcerative colitis(UC).

ABSTRACT

Objective To investigate the effect and mechanism of melatonin on bioactivity of fibroblasts from human hypertrophic scar. Methods Fibroblasts from hypertrophic scar were cultured and incubated with melatonin, melatonin and Luzindole, and Luzindole, respectively, for 24 h,and the media as control.XTT/PMS assay was used to measure the proliferation of fibroblasts,ELISA assay to detect the TGF-β1 production of fibroblasts, and the expression of cell a-SMA,collagen Ⅰ,collagen Ⅲ mRNA were determined with real-time PCR method. Results Compared with the control, melatonin at the concentration of 10-5mmol/L,10-3mmol/L,and 1 mmol/L could inhibit the proliferation of fibroblasts in a does-dependent manner (P<0.05); melatonin at the concentration of 10-3 mmol/L could significantly decrease the TGF-β1 production and expression of a-SMA mRNA and collagen Ⅰ mRNA in fibroblasts from human hypertrophic scar (P<0.05) ; the effect of melatonin on fibroblast was significantly blocked by Luzindole (P<0.05),but melatonin could not inhibit collagen Ⅲ mRNA expression (P>0.05).Conclusions Melatonin can significantly regulate the biological activity of fibroblasts from human hypertrophic scar through a receptor pathway.
